Abstract

This study aims to investigate the association of rs8192678 polymorphism in the peroxisome proliferator-activated receptor gamma coactivator 1 (PPARGC1) gene with obesity and overweight. The rs8192678 polymorphism was genotyped in all participants. In addition, a systematic review and meta-analysis of the literature for genetic association studies was conducted by searching PubMed, Scopus, and Google scholar before January 2022. The case-control study showed an increased risk for obesity in the allele model (OR: 1.45, 95%CI = 1.00–2.20, P value = 0.05). Ten case-control studies including 7821 individuals (3150 obese, 648 overweight, and 4023 normal weight) were included in the meta- analysis. Statistical analysis revealed an increased risk of overweight/obesity with polymorphism in the recessive model (OR: 1.18, 95%CI = 1.02–1.37, P value = 0.03), homozygous model (OR: 1.28, 95%CI = 1.01–1.62, P value = 0.37), and heterozygous model (OR: 1.16, 95%CI = 1.02–1.34, P value = 0.48). Our results suggest that the rs8192678 polymorphism was correlated with a higher risk of obesity in a case-control study and meta-analysis. © 2022
