Abstract

In the current study, we used electrospun cellulose acetate scaffolds as a delivery vehicle for Plerixafor and menstrual blood stem cells (MenSCs) for diabetic wound healing applications. Cellulose acetate scaffolds were incorporated with 1.5% w/w Plerixafor and then seeded with 30000 MenSCs. The developed scaffolds were studied regarding their various physicochemical and biological properties. A wound healing assay was performed in the excisional diabetic wound rat model. Results showed that the cellulose acetate/Plerixafor/MenSCs group had a significantly higher wound healing rate than other groups. This group showed 93.72 ± 6.49% of wound size reduction, 45.66 ± 5.44 μm of epithelial thickness, and 46.25 ± 6.10% of collagen deposition at the end of the 14th day post-surgery. In addition, the wound healing function of these dressings was associated with a significant increase in the tissue expression level of the vascular endothelial growth factor gene (VEGF). The cellulose acetate/Plerixafor/MenSCs group significantly reduced the NFKβ gene expression. © 2022 Elsevier B.V.
